Phosphodiesterase (PDE) Inhibition

PDE inhibitors are used clinically for erectile dysfunction (PDE5i like Viagra), weight loss, and bronchodilation/asthma treatment.

Symplocos Racemosa is one of nature’s most well-documented phosphodiesterase inhibitors. Phosphodiesterase is the enzyme responsible for breaking down cyclic-AMP (cAMP). cAMP is a second messenger that has beneficial effects like improved fat loss, increased muscle mass, improved thyroid function, and improved steroidogenesis. Indeed, the very reason forskolin works is because it increases levels of cAMP in the body by increasing levels of the enzyme that creates cyclic AMP.

A simplified model:

Enzyme A -> cAMP pool of the body -> Enzyme B

Enzyme A is creating more cAMP for the aforementioned benefits. This is the enzyme that forskolin increases, resulting in the well-documented effects on human body composition.
Enzyme B is breaking down cAMP and working against enzyme A. Enzyme B is known as phosphodiesterase (PDE). Symplocos racemosa contains PDE inhibitors, so levels of Enzyme B decrease and levels of cAMP increase. The net result is increased levels of cAMP.

The specific PDE they tested against in these studies was PDE1, but this does not mean that Lodhra bark is not active against other PDE isoforms. PDE1 inhibition is a mechanism shared by selegiline (Deprenyl), but its main documented role is enhanced pulmonary function, improved energy levels, and increased vasodilation in response to nitric oxide (Invalid Link Removed).



Hormonal Effects

Aside from increasing cAMP which increases steroidogenesis, Lodhra also increases LH/FSH (Invalid Link Removed) and was effective in treating a murine model analogous to secondary infertility. In males, LH is known to stimulate testosterone production while FSH nourishes developing sperm.


Complementary Effects with Arachidonic Acid (ArA)


ArA has three primary metabolites: prostaglandins (associated with muscle accretion and vasodilation), thromboxanes (associated with blood clotting; inhibited by carnitines or aspirin), and leukotrienes (associated with constriction of smooth muscle, such as bronchoconstriction). The latter two effects are typically considered unfavorable.
Lodhra Bark is a lipoxygenase inhibitor, preventing the conversion of ArA to leukotrienes. This leaves more ArA available to be converted to prostaglandins or thromboxanes, the latter of which can be inhibited by methods stated above.

Lodhra Bark extract has been shown to reverse markers of liver damage and even change the histology (cell structure) of the liver from a diseased state to a healthy state.

Wound Healing



Lodhra is one of few plants that has been deemed effective in the healing of wounds, both internally (ulcers, etc) and externally.

Gastrointestinal Health


Lodhra has been traditionally used as an anti-diarrheal and for preventing “looseness of the bowels.” (Invalid Link Removed). It also seems to have a curative effect on GI ulcers that is comparable to clinical drugs.
 
Yes indeed. I didn't cite them all, but it has several human trials showing normalization of menstrual periods/cycles and such, as well as helping with infertility. I didn't cite them because people get confused about studies in women, but the bottom line is the same: increased FSH and LH release. If you're not a woman, this doesn't result in increased estrogen; it results in increased testosterone. Regardless, this compound stands out to me for the large amount of data on PDE inhibition.

Sounds like clomid in the way it would help womens fertility, but interesting you said it will not increase estrogen... Why not?

This is why I didn't want to get into it, but FSH/LH increase estrogen in women. They are gonadotropins. In men, they increase our gonadal hormone: testosterone.
 
I'm on now going to take out a mortgage and buy lots of ABE.

Question Coop, as.a PDE inhibitor is there any recommendation here for use alongside nitrates/NO products?

The only reason you should worry about the cancer warning is if you've been diagnosed with it or possibly have had a family history of cancer.

my friend if you feel uncomfortable in any way leave off taking "whatever" it will in the long run make not much difference natty sups. eat train sleep...admit I have my sup. I like very much when uncomfortable on one simply I leave off taking it.

reality is tune in what you feel is right for you

and I promise with all the sups. out there you will find some indeed right for you

ArA is in natural foods we eat. Are you scared of eating those foods?

But I can understand where you are coming from. But again, if you haven't been diagnosed or have a family history, you should be fine.

It is not wrong to be cautious. ArA has so many different metabolic pathways and they are extremely complex. If someone says that it is 100% safe, I would probably think twice on trusting their opinion. That being said, I'm taking it right now. You need to decide if the juice is worth the squeeze. I always wondered this as well, even Alphamine uses a component.

C20 never seems to bring out any results though
 
God damn... I WANT IN ON THIS NOW! Running ArA at the moment though, so looks like I will run ABE and new EP together for 8 weeks, take a break for a month - or just run something else (:P) then back to the ArA stacked with ABE haha.

Can anyone here comment on the need to dose everyday vs workout days only to compliment ARA? I have c20 and am going to use it to compliment the ARA but since I workout 3 or at most 4 days/wk is it possible or even useful to only dose on workout days with the ara or does the lodhra need to build up?

I would think that if you are taking ara everday then your best bet would be to supplement the lodhra everday as well. I will ask Coop to double check that for you though

I agree, what I'm asking is do I need to take the c20 on days I don't lift/take ARA? I will only be lifting 3-4 days out of the week so if I only dose on workout days I can double the length of time I use the c20.
