GDF-8 shRNA

Well, i posted something about this on another site i visit alot and has many reputable people, especially in the AAS/Peptide catergory and got this feedabck...
If he injected himself with a viral vector to silence myostatin, he is a complete idiot. There is so many problems with this, i don't even want to get into it.

Gene therapy has been almost universally unsuccessful, requires repeated dosing and continued modification of one's genomic DNA, and has resulted in many deaths. For instance, from the induction of cancer.

I am amazed he got ahold of this. Basically, this is really only available for use in laboratory animals.

If this guy really is doing this, you need to tell hiim that it is very dangerous and it's not permanent, as he believes from the thread. Basically the vector will integrate and express for a few months and then get ****ed up by methylation and DNA excision/repair mechanisms.

It is very unfortunate if he is actually doing this. Again, this is the kind of thing that's only available for administering to laboratory animals for scientific experiment.

hmm well they are wrong in one aspect, it is available for human use, however its meant for in-vitro use. places I found allowed you to specify rat or human shRNA for a variety of things.

yes, i did mention that. Im waiting for a response and i did ask him if he'd be willing to sign up here and talk about anything. He is very reputable and experienced and i think he'd be a great addition over here.
 
You need to find out if he's using a viral vector that integrates into the host genome. It should be something like lentivirus or adenoassociated virus. There aren't a lot of other ways to do what he's purportedly doing, so it likely is a viral vector.

Next, there has really only been one successful treatment with gene therapy. Something for a liver disease. This means what he's doing hasn't made it through safety and efficacy at the FDA, which means there is no "human grade."

depends on your definition of "human grade". it is available targeted for human genetic code. part of dna therapy being unsuccessful is that a genetic cause isn't as cut and dried as it is in this particular instance. most genetic anomalies are a case of the gene not functioning properly and the therapy is designed to make the gene work correctly. in this case we are talking about silencing a specific well known gene where we have good documented cases of both the gene working normally, and the gene being silent (the human myostatin deficient cases). So him saying there is no human grade is irrelevant, using that sort of logic there never could be human grade - since if you can't produce material to run FDA testing with, how do you ever get FDA approved?

granted, this doesn't mean i'd try it :)
 
listen i have never said anything about this being approved for human use or anything of the sort if you are interested in the results i will post them im not interested in debating someone calling me and idiot this i already know .
 
Haha. That last part of the sentence threw me for a loop, Black! Dude, you're doing something that very few have done before, so you know all the "gurus" are going to have something to say. It's already been done and I for one and quite interested in hearing/seeing your results.
 
I just wanna be clear. In no way was i trying to cut BLACK. He is pioneering and puttin his body on the line for a small community of us, not sure if he is loggin this any where else or will be a future study.

I simply mentioned it on another board and a member had is opinion over there. I posted it in a neutral position on the topic.

Sorry for causing any stress on you BLACK or anyone else.

So you do it once every few weeks? How long does it last? Could one do it Once a month or twice a month?

I mean I am looking to try it as well with a stack of pegmgf and ghrp6 even if it was just once a month and that gave some results I am game.
 
DJ, just chill out and learn something. Why would you be so wiling to do something like this without having anything/one to compare it to/ learn from. Just eat, workout, rest, repeat. Let Black share his experience in peace.

DJ, thats exactly why i dint answer you. It doesnt seem you research anything.
Also, BLACK seems to know his shyt pretty well. He is doing this as prolly one of the first and currently only...why would you just go do it? Cause its easy? Quick? i want to too but i wont until there are more guys like BLACK and research is out.

I meant shyt, we are still finding things out about GHRP, Peg, IGF, etc etc...this log hasent and prolly will not even touch the 1st floor of a 1,000 story building.

PS-I also dint answer casue a simple google search brings up a major amount of info and labs.

alright sorry about not posting yesterday ate a sandwich from a restaraunt and it got me sick feel better today will get back in the gym tommorrow but all is good know just to make this clear i have never said or will i say that this is approved for human use i just have my personal conviction when it comes to myostatin and if this stuff dont work i will tell everyone it dont work but if it do i will say it does peace!

well wassup family its your man black another day in the everyday struggle against myostatin so far its been a week since i took the silencer and right know i have to say besides the absolute robust hunger all the time that i am noticing a little belly fat starting to melt away know to put this in perspective i was sick yesterday and the day before and my appetite was all screwed up so that could have something to do with the fat shedding but like my appetite is crazy right know family havent noticed any direct strength gains but we will see in the near future if anything happens
 
alright peeps wassup its your man black back again with another log well its been ten days since my log has started and so far the biggest thing im noticing is belly fat lost and it has been nice losing fat while having and appetite from h###!
my weight right know is at 230 lbs but i managed to get my waist down from a 36 to a 34 and part of the reason my belly fat was at a 36 anyway is because i ate any and everything and i do mean everything even when i was doing the anti-bodies which i did not do it any justice my main goal on that was strength and for that reason i did not concentrate on cardio at all but i must say it was because of the myostatin anti-bodies that i was not and obese man anyway enough of my mouth oh i know i was supposed to post pic but rest assure they are coming peace!

This is a good sign, you've gained some muscle and dropped some fat :)
 
So if you see this insanely huge / ripped black man walking down the street sporting a pair of extra traps that pop out of his normal traps, you'll know who it is!
 
alright sorry about the delay in posting been busy and so far the hungerness is still there the tiredness is not the bad anymore but the fat burning has been great i havent notice any strength gains at all since i began this experiment so its gets and F on the front other than that my weight right know is 228 pounds oh i must add this the pumps i get in the gym is outstanding anyway i will be able to post better next week and show some before and after shots any question feel free to ask.
 
well the experiment unfortunate is yeilding less than desirable results although i remain strong there have been little noticeable muscle growth i remain optimistic on future experiments but this one here seems to be a failure although the first two weeks fatloss was amazing but since have been null and void im gonna give it another week then im closing this thread down one thing good is that i have not grown and extra arm or worst!

maybe the mysotatin was only 'silenced' for the first 2 weeks.
perhaps either the dosage wasnt correct or the sequence is slightly out? im just guessing but if your myostatin was truelly non existant im sure we would be seeing something extra. that said it could take months to see the difference, 2-3 weeks just isnt long enough bro.

anyway good on you for trying this out, the only reservation i have is when i read the study that suggested mysotatin maybe involved in tendon integrity as a positive regulator.
and as you know with this pec repair ive just gone through, the last thing i could risk is weakening my tendons, especially since i now have some unknown person's 2 hamstring tendons holding my left pec onto the humerous bone!:aargh:
 
thats true one thing though my strength is still up i mean just yesterday i was able to get 315 for 4 sets of 15 on the bench not bad rightjust that muscle growth is not there and also i got to add that im not using anything else at this time i got a feeling on what went wrong but im not willing to try it again at this time to risky right know.
 
How can you say there is not muscle growth if within a period of a few weeks you maintained weight but drastically reduced bodyfat?

Not to hijack, but just wanted to say Pumb, that it's a medical miracle that we're able to take someone else's hamstring tendons and use them to repair you. That's incredible, so please don't be TOO discouraged. :D I hope you're all good to go and America has treated you well.

Big Boy Black, thank you for posting your experiences. I agree with Pumb that results will take longer than 2 weeks. The fat loss in the 1st 2 weeks is indicative of SOMETHING happening and muscle growth takes longer than fat loss dood. Furthermore, your splicer knocks out a gene (myostatin), so that means for other cells to appreciate the now missing gene, the cells have to reproduce (replicate) and that takes time. Hang in there biggie!

that is correct maybe i do need to chill out a bit and wait it out more patience is key when you are dealing with this stuff
 
im here man been going threw personal issues with the old lady everything is the same my weight is stuck at 228 my strength is still good 435 on the bench 675 on squats although my knee has been bothering me lately so im gonna slack off of the extremely heavy squats for a little while other than that there has been a slight increase in trap size no increase in leg mass a little more back detail look im not using nothing else currently and im not consuming nearly as many calories as i was when i was on the anti-bodies so this stuff has help me maitain weight and strenght even during a strict diet or calorie consumption

hey wassup papa everythings everything you know me and the wifey getting along better just had to refocused if you know what i mean anyway the gains with this stuff have come to a hault when i first took this stuff it was a couple of things i guess i should have done but i didnt but thats another story the first 2 weeks on this stuff the fulness efeect was out of this world and also the fat reduction but since then it has been dwindling i guess more research needs to be done as i believe alot of promise is in this area. and i intend to explore it.my next log will be on this new joint supplement and preworkout drink from my homeboy king stay tuned for that after that its right back at the research area with a detailed log on GW501516 so black is back people
