Capsaicin is a transient receptor potential vanilloid 1 (TRPV1) agonist. TRPV1 is a Ca2+ permeant non-selective cation channel expressed in a subpopulation of primary afferent neurons, conducting impulses inwards to the brain. It is included in TR1UMPH for its ability to induce increases in [Ca2+]. It has been established that the activation of mTOR and subsequent muscle hypertrophy requires a (TRPV1) mediated increase in intracellular calcium concentrations ([Ca2+]I). Thus, calcium signaling is a crucial step for mechanical load-induced activation of mTOR.
In an animal model, muscle cells were treated with the TRPV1 agonist capsaicin to analyze the effect on [Ca2+] using Fluo-4. [Ca2+] was increased by capsaicin in wild-type but not in TRPV1-null muscle fibers, indicating that the capsaicin induced increases in [Ca2+]i were mediated by TRPV1 (Fig.1A and 1B). Since [Ca2+] regulates divergent intracellular signaling pathways, the capsaicin-induced intracellular signaling events were further analyzed. Similar to previous observations, intramuscular injection of capsaicin induced phosphorylation of p70S6K at Thr389, an mTOR- regulated phosphorylation site that is necessary for p70S6K activation in addition to phosphorylation of the downstream target S6. Capsaicin also induced phosphorylation of p70S6K at Thr421/Ser424 and mTOR at Ser2448 and Ser2481. In addition to p70S6K, capsaicin induced phosphorylation of Erk1/2 and p38 MAPK, but not Akt, AMPKα, or GSK3β.

Capsaicin can also improve lipolysis which is the decomposition of stored fat. This process activates enzymes that increase production of Cyclic adenosine monophosphate (cAMP) which activates protein kinase A.
In a study with 10 healthy males, ingestion of 150mg of capsaicin ingestion (150mg) enhances substrate oxidation associated with thermogenic sympathetic activity as an energy metabolic modulator without causing prolongation of the cardiac OT interval during aerobic exercise in humans. In conclusion, it may be considered that capsaicin consumption 1 h before low intensity exercise (50% VT(max)) is a valuable supplement for the treatment of individuals with hyperlipidemia and/or obesity because it improves lipolysis without any adverse effects on the cardiac depolarization and repolarization process.
Prevention of muscle atrophy is based on either inhibiting protein-degradation pathways, which are activated during atrophy, or activating protein-synthesis pathways, which induce muscle hypertrophy.. As we previously reviewed, Capsaicin achieves the latter. Rhodiola Rosea Extract, a Scandinavian herb, possesses a significant ability to reduce stress on the body, particularly exhaustion and fatigue, caused through physical exertion. Rhodiola Rosea has also been shown to possess antioxidant properties which will also moderate undue stress to your body from intense exercise.
24 healthy volunteers, 12 male and 12 female, were given a dose of 100mg Rhodiola Rosea. After just 4 days of supplementation a significant increase in time to exhaustion, VO2, VCO2, peak O2 output, and peak CO2 output was observed.
Another study, with 22 professional rowers, that were given 100mg of Rhodiola Rosea twice daily for a 4 week period found a significant increase in total plasma antioxidant capacity and a decrease in superoxide dismutase (SOD) activity. SOD is an enzyme produced as a result of oxygen metabolism (ie. intensive activity) and can lead to cell damage if not mitigated. These results suggest that continuous supplementation of Rhodiola Rosea can improve endurance.

BMPs include a large family of proteins within the TGF-beta superfamily of growth and differentiation factors. Members of the BMP family have been found to be key signaling molecules in embryogenesis, in species ranging from small flies to humans. Research has shown BMP signaling is the fundamental signal for hypertrophy, specifically BMP7 which is the pathway that has the most influence on muscle growth. BMP7 can stimulate osteoblast differentiation leading to formation of skeletal muscle. Unfortunately the majority research on Salidroside is limited to testing on mice. However, given the positive human data on Rhodiola Rosea and the preclinical data in humans it lends extra credibility to these findings.

An ergogenic aid can be broadly defined as a substance used for the purpose of enhancing athletic performance. There are several ergogenic aids that are so beneficial for high intensity exercise that they are considered “staples”. We included three such staples in TR1UMPH to elicit an increase in physical capacity and reduction in fatigue; 2.5g of creatine Hcl, 2.5g of betaine, and 2g of L-Carnitine L-Tartrate (LCLT).
Creatine is by far the most popular ergogenic supplement because it has proven to be effective by numerous studies. In fact, it is likely the most studied performance enhancing supplement in the industry. Of course we cannot review all of these studies so in order to be efficient; let's review a meta-analysis.
One meta-analysis reviewed 16 studies involving healthy male and/or female subjects. The summary difference in maximum weight lifted was 6.85kg (95% confidence interval [CI], 5.24-8.47) greater after creatine than placebo for bench press and 9.76 kg (95% CI, 3.37-16.15) greater for squats; there was no difference for arm curls. In 7 of 10 studies evaluating maximal weight lifted in young men (younger than 36 years) that engaged in resistance training there was no difference in cycle ergometer or isokinetic dynamometer performance. The researchers concluded oral creatine supplementation combined with resistance training increases maximal weight lifted in young men. There is no evidence for improved performance in older individuals or women or for other types of strength and power exercises
A second meta-analysis of 96 English-language, peer-reviewed papers (100 studies), which included randomized group formation, a placebo control, and human subjects who were blinded to treatments was performed. It compared the effects of creatine supplementation (CS) on effect size (ES) for body composition (BC) variables (mass and lean body mass), duration and intensity (< or = 30 s, [ATP-PCr = A]; 30-150 s [glycolysis = G]; >150 s, [oxidative phosphorylation = O]) of the exercise task, type of exercise task (single, repetitive, laboratory, field, upper-body, lower-body), CS duration (loading, maintenance), and subject characteristics (gender, training status). ES was greater for changes in lean body mass following short-term CS, repetitive-bout laboratory-based exercise tasks < or = 30 s (e.g., isometric, isokinetic, and isotonic resistance exercise), and upper-body exercise. There is no evidence in the literature of an effect of gender or training status on ES following CS
TR1UMPH uses creatine hydrochloride (HCL) which is creatine bound with hydrochloric acid. It’s turned into a basic creatine molecule by stomach acid so you get all the benefits of creatine without any potential side effects like upset stomach. Creatine HCL is also more acidic (ie. lower PH) than creatine monohydrate which makes it more soluble and would suggest an increase in absorption. Typically the pH in the upper gastrointestinal tract of healthy males and females is often low enough to counteract the neutral pH of creatine monohydrate (ie. pH ~7). However, research shows after the administration of a solid and liquid meal gastric pH reached a peak of 6.7 pH and then declined gradually back to a fasted state after approximately two hours. This data suggests supplementing with creatine monohydrate in a fed state could affect absorption. Creatine HCL is a suitable alternative due to its lower pH value.
Trimethylglycine (TMG), typically referred to as betaine anhydrous, is derived from the beetroot and is an active metabolite of choline in the body. It serves a vital role in methylation in the body, particularly of the amino acid homocysteine to form methionine. Methionine contributes to the synthesis of creatine in the body. Therefore supplementing with betaine will lead to an increase in muscle strength through its ability to increase creatine production in the body. It also been found to increase protein synthesis.
One study, involving 12 trained men (age 19.7 ± 1.23 years), examined the effect of betaine supplementation on selected circulating hormonal measures and Akt muscle signaling proteins after an acute exercise session over the course of 2 weeks. Betaine vs. placebo supplementation enhanced both the anabolic endocrine profile and the corresponding anabolic signaling environment, suggesting increased protein synthesis.
Twenty-three experienced strength trained men, matched for training experience (4.8 ± 2.3 years) and body fat percentage (BF%: 16.9 ± 8.0%) were randomly assigned to either a placebo (PL; n = 12) or betaine group (BET; n = 11; 2.5 g/day), and completed a 6 week periodized training program to assess the effect of long term betaine supplementation on body composition, performance, and homocysteine thiolactone (HCTL). Arm CSA increased significantly (p < .05) in BET but not PL. No differences existed between group and time for changes in thigh CSA. Back squat training volume increased significantly (p < .05) for both groups throughout training. Bench press training volume was significantly (p < .05) improved for BET compared to PL at microcycles one and three. Body composition (BF%, FM, LBM) improved significantly (p < .05) in BET but not PL. A significant interaction (p < .05) existed for HCTL, with increases from BL to week 2 in PL, but not BET. Additionally, HCTL remained elevated at week 4 in PL, but not BET. The study concluded that six-weeks of betaine supplementation improved body composition, arm size, bench press work capacity, attenuated the rise in urinary HCTL, and tended to improve power.

L-carnitine is synthesized from the amino acids lysine and methionine and performs essential functions in human metabolism. L-carnitine supplementation has been found to have a beneficial effect on markers of postexercise metabolic stress and muscle damage.
A study involving eight healthy males examined the required dose of l-carnitine to elicit a reduction in postexercise metabolic stress and muscle damage. The study participants were given doses of 0g, 1g, and 2g of LCLT for 3 weeks and then performed a bout of resistance exercise (5 sets of 15-20 repetition maximum with a 2-min rest between sets) with associated blood draws. Serum carnitine concentrations increased (p < or = 0.05) following the 1g and 2g doses, with the 2-g dose providing the highest carnitine concentrations. The 1g and 2g doses reduced postexercise serum hypoxanthine, serum xanthine oxidase, serum myoglobin, and perceived muscle soreness. In conclusion, both the 1g and 2g doses were effective in mediating various markers of metabolic stress and of muscle soreness. Use of LCLT appears to attenuate metabolic stress and the hypoxic chain of events leading to muscle damage after exercise.
18 healthy and active subjects (9 female and 9 male) participated in a double-blind, placebo, balanced treatment presentation and crossover design where they ingested 2 g/d of l-carnitine for 24 days to examine markers of performance and recovery from physical exertion. Two grams of l-carnitine supplementation had positive effects and significantly (P < or = .05) attenuated biochemical markers of purine metabolism, free radical formation, muscle tissue disruption, and muscle soreness after physical exertion

Measurements were taken 30 minutes post-exercise and then the subjects performed three sets of bench press throws at 50% of their predetermined 1-repetition maximum to assess peak force, peak power, and rate of force development. All trials were performed after an overnight fast, a 48-hour abstention from intense exercise, and during the same time of day to minimize diurnal variation. Compared to baseline (pre) values, peak GH increased 44-fold during A-GPC (from 0.19 ± 0.06 to 8.4 ± 2.1 ng/mL) vs. 2.6-fold during placebo (from 1.9 ± 0.8 to 5.0 ± 4.8 ng/mL, P < 0.03) (Figure 3). Peak bench press force was 14% greater in A-GPC (933 ± 89 N) vs. placebo (818 ± 77 N, P < 0.02).

L-glutamine is an amino acid that comprises more than 60% of skeletal tissue. Supplementation with l-glutamine is believed to reduce recovery time and aid in protein synthesis. Glutamine has been shown to inhibit protein breakdown in rat skeletal muscle. However, a human study conducted seems to refute that assertion. TRPV1 is a Ca2+ permeant non-selective cation channel expressed in a subpopulation of primary afferent neurons, conducting impulses inwards to the brain. It is included in TR1UMPH for its ability to induce increases in [Ca2+]. It has been established that the activation of mTOR and subsequent muscle hypertrophy requires a (TRPV1) mediated increase in intracellular calcium concentrations ([Ca2+]I). Thus, calcium signaling is a crucial step for mechanical load-induced activation of mTOR.
In an animal model, muscle cells were treated with the TRPV1 agonist capsaicin to analyze the effect on [Ca2+] using Fluo-4. [Ca2+] was increased by capsaicin in wild-type but not in TRPV1-null muscle fibers, indicating that the capsaicin induced increases in [Ca2+]i were mediated by TRPV1 (Fig.1A and 1B). Since [Ca2+] regulates divergent intracellular signaling pathways, the capsaicin-induced intracellular signaling events were further analyzed. Similar to previous observations, intramuscular injection of capsaicin induced phosphorylation of p70S6K at Thr389, an mTOR- regulated phosphorylation site that is necessary for p70S6K activation in addition to phosphorylation of the downstream target S6. Capsaicin also induced phosphorylation of p70S6K at Thr421/Ser424 and mTOR at Ser2448 and Ser2481. In addition to p70S6K, capsaicin induced phosphorylation of Erk1/2 and p38 MAPK, but not Akt, AMPKα, or GSK3β.

Capsaicin can also improve lipolysis which is the decomposition of stored fat. This process activates enzymes that increase production of Cyclic adenosine monophosphate (cAMP) which activates protein kinase A.
In a study with 10 healthy males, ingestion of 150mg of capsaicin ingestion (150mg) enhances substrate oxidation associated with thermogenic sympathetic activity as an energy metabolic modulator without causing prolongation of the cardiac OT interval during aerobic exercise in humans. In conclusion, it may be considered that capsaicin consumption 1 h before low intensity exercise (50% VT(max)) is a valuable supplement for the treatment of individuals with hyperlipidemia and/or obesity because it improves lipolysis without any adverse effects on the cardiac depolarization and repolarization process.
Prevention of muscle atrophy is based on either inhibiting protein-degradation pathways, which are activated during atrophy, or activating protein-synthesis pathways, which induce muscle hypertrophy.. As we previously reviewed, Capsaicin achieves the latter. Rhodiola Rosea Extract, a Scandinavian herb, possesses a significant ability to reduce stress on the body, particularly exhaustion and fatigue, caused through physical exertion. Rhodiola Rosea has also been shown to possess antioxidant properties which will also moderate undue stress to your body from intense exercise.
24 healthy volunteers, 12 male and 12 female, were given a dose of 100mg Rhodiola Rosea. After just 4 days of supplementation a significant increase in time to exhaustion, VO2, VCO2, peak O2 output, and peak CO2 output was observed.
Another study, with 22 professional rowers, that were given 100mg of Rhodiola Rosea twice daily for a 4 week period found a significant increase in total plasma antioxidant capacity and a decrease in superoxide dismutase (SOD) activity. SOD is an enzyme produced as a result of oxygen metabolism (ie. intensive activity) and can lead to cell damage if not mitigated. These results suggest that continuous supplementation of Rhodiola Rosea can improve endurance.
As previously mentioned TR1UMPH contains a purer extract of Rhodiola Rosea which yields 11% Salidroside. That is no coincidence! BMPs include a large family of proteins within the TGF-beta superfamily of growth and differentiation factors. Research has shown BMP signaling is the fundamental signal for hypertrophy, specifically BMP7 which is the pathway that has the most influence on muscle growth. BMP7 can stimulate osteoblast differentiation leading to formation of skeletal muscle. Unfortunately the majority research on Salidroside is limited to testing on mice. However, given the positive human data on Rhodiola Rosea and the preclinical data in humans it lends extra credibility to these findings.

An ergogenic aid can be broadly defined as a substance used for the purpose of enhancing athletic performance. There are several ergogenic aids that are so beneficial for high intensity exercise that they are considered “staples”. We included three such staples in TR1UMPH to elicit an increase in physical capacity and reduction in fatigue; 2.5g of creatine Hcl, 2.5g of betaine, and 2g of L-Carnitine L-Tartrate (LCLT).
Creatine is by far the most popular ergogenic supplement because it has proven to be effective by numerous studies. In fact, it is likely the most studied performance enhancing supplement in the industry. Of course we cannot review all of these studies so in order to be efficient; let's review a meta-analysis.
One meta-analysis reviewed 16 studies involving healthy male and/or female subjects. The summary difference in maximum weight lifted was 6.85kg (95% confidence interval [CI], 5.24-8.47) greater after creatine than placebo for bench press and 9.76 kg (95% CI, 3.37-16.15) greater for squats; there was no difference for arm curls. In 7 of 10 studies evaluating maximal weight lifted in young men (younger than 36 years) that engaged in resistance training there was no difference in cycle ergometer or isokinetic dynamometer performance. The researchers concluded oral creatine supplementation combined with resistance training increases maximal weight lifted in young men. There is no evidence for improved performance in older individuals or women or for other types of strength and power exercises
A second meta-analysis of 96 English-language, peer-reviewed papers (100 studies), which included randomized group formation, a placebo control, and human subjects who were blinded to treatments was performed. It compared the effects of creatine supplementation (CS) on effect size (ES) for body composition (BC) variables (mass and lean body mass), duration and intensity (< or = 30 s, [ATP-PCr = A]; 30-150 s [glycolysis = G]; >150 s, [oxidative phosphorylation = O]) of the exercise task, type of exercise task (single, repetitive, laboratory, field, upper-body, lower-body), CS duration (loading, maintenance), and subject characteristics (gender, training status). ES was greater for changes in lean body mass following short-term CS, repetitive-bout laboratory-based exercise tasks < or = 30 s (e.g., isometric, isokinetic, and isotonic resistance exercise), and upper-body exercise. There is no evidence in the literature of an effect of gender or training status on ES following CS
TR1UMPH uses creatine hydrochloride (HCL) which is creatine bound with hydrochloric acid. It’s turned into a basic creatine molecule by stomach acid so you get all the benefits of creatine without any potential side effects like upset stomach. Creatine HCL is also more acidic (ie. lower PH) than creatine monohydrate which makes it more soluble and would suggest an increase in absorption. Typically the pH in the upper gastrointestinal tract of healthy males and females is often low enough to counteract the neutral pH of creatine monohydrate (ie. pH ~7). However, research shows after the administration of a solid and liquid meal gastric pH reached a peak of 6.7 pH and then declined gradually back to a fasted state after approximately two hours. This data suggests supplementing with creatine monohydrate in a fed state could affect absorption. Creatine HCL is a suitable alternative due to its lower pH value.
Trimethylglycine (TMG), typically referred to as betaine anhydrous, is derived from the beetroot and is an active metabolite of choline in the body. It serves a vital role in methylation in the body, particularly of the amino acid homocysteine to form methionine. Methionine contributes to the synthesis of creatine in the body. Therefore supplementing with betaine will lead to an increase in muscle strength through its ability to increase creatine production in the body. It also been found to increase protein synthesis.
One study, involving 12 trained men (age 19.7 ± 1.23 years), examined the effect of betaine supplementation on selected circulating hormonal measures and Akt muscle signaling proteins after an acute exercise session over the course of 2 weeks. Betaine vs. placebo supplementation enhanced both the anabolic endocrine profile and the corresponding anabolic signaling environment, suggesting increased protein synthesis.
Twenty-three experienced strength trained men, matched for training experience (4.8 ± 2.3 years) and body fat percentage (BF%: 16.9 ± 8.0%) were randomly assigned to either a placebo (PL; n = 12) or betaine group (BET; n = 11; 2.5 g/day), and completed a 6 week periodized training program to assess the effect of long term betaine supplementation on body composition, performance, and homocysteine thiolactone (HCTL). Arm CSA increased significantly (p < .05) in BET but not PL. No differences existed between group and time for changes in thigh CSA. Back squat training volume increased significantly (p < .05) for both groups throughout training. Bench press training volume was significantly (p < .05) improved for BET compared to PL at microcycles one and three. Body composition (BF%, FM, LBM) improved significantly (p < .05) in BET but not PL. A significant interaction (p < .05) existed for HCTL, with increases from BL to week 2 in PL, but not BET. Additionally, HCTL remained elevated at week 4 in PL, but not BET. The study concluded that six-weeks of betaine supplementation improved body composition, arm size, bench press work capacity, attenuated the rise in urinary HCTL, and tended to improve power.

L-carnitine is synthesized from the amino acids lysine and methionine and performs essential functions in human metabolism. L-carnitine supplementation has been found to have a beneficial effect on markers of postexercise metabolic stress and muscle damage.
A study involving eight healthy males examined the required dose of l-carnitine to elicit a reduction in postexercise metabolic stress and muscle damage. The study participants were given doses of 0g, 1g, and 2g of LCLT for 3 weeks and then performed a bout of resistance exercise (5 sets of 15-20 repetition maximum with a 2-min rest between sets) with associated blood draws. Serum carnitine concentrations increased (p < or = 0.05) following the 1g and 2g doses, with the 2-g dose providing the highest carnitine concentrations. The 1g and 2g doses reduced postexercise serum hypoxanthine, serum xanthine oxidase, serum myoglobin, and perceived muscle soreness. In conclusion, both the 1g and 2g doses were effective in mediating various markers of metabolic stress and of muscle soreness. Use of LCLT appears to attenuate metabolic stress and the hypoxic chain of events leading to muscle damage after exercise.
18 healthy and active subjects (9 female and 9 male) participated in a double-blind, placebo, balanced treatment presentation and crossover design where they ingested 2 g/d of l-carnitine for 24 days to examine markers of performance and recovery from physical exertion. Two grams of l-carnitine supplementation had positive effects and significantly (P < or = .05) attenuated biochemical markers of purine metabolism, free radical formation, muscle tissue disruption, and muscle soreness after physical exertion

All trials were performed after an overnight fast, a 48-hour abstention from intense exercise, and during the same time of day to minimize diurnal variation. Compared to baseline (pre) values, peak GH increased 44-fold during A-GPC (from 0.19 ± 0.06 to 8.4 ± 2.1 ng/mL) vs. 2.6-fold during placebo (from 1.9 ± 0.8 to 5.0 ± 4.8 ng/mL, P < 0.03) (Figure 3). Peak bench press force was 14% greater in A-GPC (933 ± 89 N) vs. placebo (818 ± 77 N, P < 0.02).

L-glutamine is an amino acid that comprises more than 60% of skeletal tissue. Supplementation with l-glutamine is believed to reduce recovery time and aid in protein synthesis. Glutamine has been shown to inhibit protein breakdown in rat skeletal muscle. However, a human study conducted seems to refute that assertion.
