regarding this protocol

running HCG week 4-12 (till the end of a cycle)

250iu twice a week (M/Th)

WOULD IT MAKE A DIFFERENCE IF used ONCE a week at 500iu, opposed to twice a week at 250iu...?

Yes, because of the half-life of hCG. 2x/week will keep it in your system for more of the week, whereas with 1x/week dosing you will have cleared most of it by day 5-6.

id also like to point out that it will lose potentcy over time. after about 4 weeks i had to switch from 250iu 2x week to ~300iu 2x week

Desensitization to hCG is still an unclear topic; I've chatted with people that ran hCG as monotherapy at several thousans IU's/week and didn't notice decreased responsiveness, then there's cases like yours. Theoretically there should be some desensitization, but it's hard to build a consensus on how much and how fast.
 
Allot is individual as well.. I have played around with dosing(250 eod, 250 e3d, e4d, 500 e3d, etc) I find i feel best dosing 500 every 3rd then 4th day(mon/thur)..

Nos- i feel you bro, for me it seems right around 3wks, as soon as i crack a new one, it is a NOTICEABLE difference.. Next time i am ordering all 1500iu's. It is more pricey, but IMO worth it.

no i mean the actual mixture HCG/BAC water lost potentcy. its said (but i think bro science) that HCG mixed with BAC water should last up to 60 days. well at about 40 days i noticed 250iu 2x week wasnt cutting it

It isn't desensitization, it is the product degrading.. If it were a case of you being desensitized then switching to a new batch, would not yield better results, and it does(at least for me)
