"The damaging evidence from many early rat studies showed severely toxic effects, including the development of cancer in the liver, uterus, or testes upon tamoxifen administration. (30-34,41) However, this evidence was largely disregarded by further test tube studies on human cell-lines which appeared to show a lack of toxic effects. (21)"

"Liver tumors were found in four out of five rats at the highest dose level of tamoxifen (45 mg/kg per day) after 52 weeks of dosing, and these appeared to be hepatocellular carcinomas in three rats."

"In summary, in vitro studies with human tissues or cells do not show significant DNA adduct formation by tamoxifen in either hepatocytes or endometrium, and binding by {alpha}-hydroxytamoxifen occurs only at high concentration, probably non-enzymically. In vivo, there is no evidence for adduct formation in human liver, but differing results have been obtained with endometrium."
