    Very interesting.

"The damaging evidence from many early rat studies showed severely toxic effects, including the development of cancer in the liver, uterus, or testes upon tamoxifen administration. (30-34,41) However, this evidence was largely disregarded by further test tube studies on human cell-lines which appeared to show a lack of toxic effects. (21)"

    30. Tamoxifen induces hepatocellular carcinoma in rat liver: a 1-year study with two antiestrogens.
Hirsimaki P, Hirsimaki Y, Nieminen L, et al.
    Arch Toxicol. 1993; 67: 49–4

"Liver tumors were found in four out of five rats at the highest dose level of tamoxifen (45 mg/kg per day) after 52 weeks of dosing, and these appeared to be hepatocellular carcinomas in three rats."

    45mg/kg/52weeks = 4.05g/200lb/52weeks
    21. Understanding the genotoxicity of tamoxifen?
    David H. Phillips
    Carcinogenesis, Jun 2001; 22: 839 - 849.

"In summary, in vitro studies with human tissues or cells do not show significant DNA adduct formation by tamoxifen in either hepatocytes or endometrium, and binding by {alpha}-hydroxytamoxifen occurs only at high concentration, probably non-enzymically. In vivo, there is no evidence for adduct formation in human liver, but differing results have been obtained with endometrium."
