One thing that's already being tested in clinical vaccine trials for cancer and HIV are DNA plasmids encoding cytokines that boost immunity. The idea is you get local deposition into the skeletal muscle and slow release of the encoded proteins. You can google "IL-12 plasmid" for instance. Think of the possibilities.
