Saccharomyces Cerevisiae (Sake Yeast)
Sake yeast is a type of yeast used to produce Japanese rice wine (Sake) and other traditional japanese foods. Unlike other types of yeast, sake yeast is enriched with adenosine analogues. Recent research has identified that adenosine acts as a homeostatic regulator of sleep and to be a link between the humoral and neural mechanisms of sleep-wake regulation. Moreover, research on the adenosine pathway in sake yeast found that it can induce deeper sleep, increase growth hormone secretion and reduce fatigue upon waking.
It was previously discovered that Japanese sake yeast enriched in adenosine analogues activates A2a receptors in vitro and induces sleep in mice. Therefore, a clinical study was undertaken to determine whether that result held in a cultured human cell line. 68 healthy participated in a double-blind placebo-controlled crossover clinical study, ingested tablets containing either 500 mg of sake yeast powder or a placebo (cellulose) 1 hour before sleep for 4 days. Electroencephalogram analyses revealed that sake yeast supplementation significantly (P = 0.03) increased delta power during the first cycle of slow-wave sleep by 110%, without changing other sleep parameters. Sake yeast supplementation also significantly increased growth hormone secretion in the urine on awakening by 137% from 3.17 ± 0.41 (placebo) to 4.33 ± 0.62 (sake yeast) pg mg(-1) creatinine (P = 0.03). Subjective sleepiness (P = 0.02) and fatigue (P = 0.06) in the morning were improved by sake yeast. Given these benefits and the absence of adverse effects during the study period, it was concluded that sake yeast supplementation is an effective and safe way to support daily high-quality, deep sleep.
Dioscin is a glucosid saponin found in plants. It has shown lipid-lowering, anti-cancer and hepatoprotective properties. More interestingly, dioscin has shown ability to stimulate growth hormone release in pituitary cells. This finding is significant because it means that AMNES1A goes well beyond inducing a deep, restful sleep. Dioscin, via GH release, will help you build lean mass and improve recovery.
Bioassay-guided fractionation of MeOH extract from fenugreek (Trigonella foenum-graecum L.) seeds resulted in the isolation of two growth-hormone release stimulators in vitro, fenugreek saponin I (1) and dioscin (9), along with two new, i.e., 2 and 3, and five known analogues, i.e., 4-8. The structures of the new steroidal saponins, fenugreek saponins I, II, and III (1-3, resp.), were determined as gitogenin 3-O-beta-D-xylopyranosyl-(1-->6)-beta-D-glucopyranoside, sarsasapogenin 3-O-beta-D-xylopyranosyl-(1-->6)-beta-D-glucopyranoside, and gitogenin 3-O-alpha-L-rhamnopyranosyl-(1-->2)-beta-D-glucopyranoside, respectively. Fenugreek saponin I (1) and dioscin (9) caused ca. 12.5- and 17.7-fold stimulation of release, respectively, of growth hormone from pituitary cells, whereas gitogenin (5) showed moderate activity. To our knowledge, this is the first study to demonstrate that saponins stimulate growth-hormone release in pituitary cells.
Kanna is a herb that is commonly referred to as Sceletium tortuosum that has been traditionally used for its cognitive effects. It has been used as a clinical treatment for anxiety and depression since it promotes a sense of wellbeing and can relieve stress. Specifically, Sceletium tortuosum works by dual 5-HT reuptake inhibition and PDE4 (phosphodiesterase subtype 4) inhibition which plays a crucial role in regulating cognition via the PDE-4-cAMP cascade signaling pathway involving phosphorylated cAMP response element binding protein (CREB). In addition to improving cognitive ability, Kanna is known to improve mood and sleep. A dose of 25mg of Sceletium tortuosum is generally well tolerated in healthy adults taken once daily for extended periods of time so there is no need to cycle off AMNES1A.
Passionflower (Passiflora incarnata) is a herb traditionally used in the Americas and Europe as a calming herb for anxiety and insomnia. Research indicates passionflower works by increasing levels of a chemical called gamma aminobutyric acid (GABA) in the brain which lowers the activity of some brain cells resulting in a relaxed feeling.
Sixty patients were randomized into two groups to receive either oral Passiflora incarnata (500 mg) (n = 30) or placebo (n = 30) as premedication, 90 min before surgery. A numerical rating scale (NRS) was used for each patient to assess anxiety and sedation before, and 10, 30, 60, and 90 min after premedication. Psychomotor function was assessed with the Trieger Dot Test and the Digit-Symbol Substitution Test at arrival in the operating room, 30 and 90 min after tracheal extubation. The demographic characteristics of patients, ASA physical status, duration of surgery, basal NRS score, sedation at the preset time intervals, and discharge time were similar in the two groups. The numerical rating scale (NRS) anxiety scores were significantly lower in the passiflora group than in the control group (P < 0.001). There were no significant differences in psychological variables in the postanesthesia care unit and recovery of psychomotor function was comparable in both groups. The study concluded that administration of oral Passiflora incarnata as a premedication reduces anxiety without inducing sedation.
Valeriana officinalis, also referred to as valerian root, is a perennial herb comprised of three essential oils; valerenic acid and valenol, valepotriate and a few alkaloids. Valerenic acid can bind to gamma hydroxybutyric acid (GABA) (A) receptors and elicit a sedative effect. It has traditionally been used to treat restlessness, tremors and anxiety. However, it is becoming increasingly popular as a sleep aid and a treatment for insomnia. The main benefit of Valeriana is its ability to improve sleep latency (time to fall asleep). Several sleep studies have concluded that a dose of 400 or 450 mg provided such improvements and higher doses (900mg) did not provide significantly better results than 450 mg.
A clinical trial was performed with cancer patients, since they are prone to sleep disorders, to evaluate the efficacy of a supplement for sleep. 119 Participants were randomized to receive 450 mg of valerian-or placebo orally 1 hour before bedtime for 8 weeks. Several fatigue endpoints, as measured by the Brief Fatigue Inventory (BFI), and the Profile of Mood States (POMS), were significantly better for those taking valerian over placebo. Participants also reported less trouble with sleep and less drowsiness on valerian than placebo. This study found Valerian officinalis can improve factors associated with sleep, including mood, drowsiness and fatigue.
A group of 8 volunteers suffering from mild insomnia participated in a double-blind, repeated measures, random-order design trial where they received a placebo, 450 mg or 900 mg of an aqueous extract of valerian root. Subjective sleep ratings were assessed by questionnaire and movements were recorded throughout the night with wrist-worn activity meters. Using the first period of 5 consecutive minutes without movement as a criterion of sleep onset, there was a significant decrease in sleep latency with 450 mg valerian compared to placebo (15.8 ± 2.2 min vs 9.0 ± 1.5 min; paired “t” = 3.37;p < 0.01). The higher dose of valerian produced no further improvement in sleep latency.This study demonstrates that Valerian root can significant decrease sleep latency with 450mg being the most effective dose to achieve that outcome.
A meta-analysis of 18 randomized clinical trials (RCTs) of Valerian preparations compared with a placebo on Medline, the Cochrane Library, Embase and Biosis were reviewed. Sleep-quality improvement quantified through visual analogical scales (SQS) and the latency time (LT) in minutes until getting to sleep were the key parametrics were analyzed. Across the studies reviewed there was clear qualitative data indicating that valerian would be effective for improvement of insomnia.
Melissa officinalis L.
Melissa officinalis also referred to as lemon balm) is a traditional herbal medicine. It has been used as a mild sedative, treatment for muscle spasms and a antibacterial agent. More recent clinical studies show Melissa officinalis can improve mood, cognitive performance, reduce stress and also may induce anxiolytic effects.
A randomized, placebo-controlled, double-blind, balanced-crossover study investigated the acute effects on cognition and mood of a standardised extract of Melissa officinalis. Twenty healthy, young participants received single doses of 300, 600 and 900 mg of Melissa officinalis or a placebo at 7-day intervals. Measures of self-rated "calmness," as assessed by Bond-Lader mood scales, were elevated at the earliest time points by the lowest dose. Acetylcholine receptor (nicotinic and muscarinic) binding were both found to be low in comparison to the levels found in previous studies which indicates a sedative effect.
L-theanine Is an amino acid that can be found in green tea. It has been known to produce mental relaxation, lower blood pressure and improve learning ability in human beings. Other research in animals have shown l-theanine decreases serotonin and increases norepinephrine in the brain. It is known to cause a relaxing feeling yet not being sedating which is the reason it is often used with stimulants such as caffeine as it can 'take the edge off'.
A study was conducted to evaluate the effect of L-theanine on the release of brain alpha waves known to be related with mental relaxation and concentration. Twenty healthy male volunteers aged 18 to 30 years without any physical and psychological diseases participated. Repeated ANOVA revealed that there were significant differences of occipital alpha power values between placebo and test groups with high anxiety (p<0.05). But there were no significant differences of frontal and occipital alpha power values between placebo and test groups with low anxiety (p>0.05) . The results of this study suggest that L-theanine containing tablets promote the release of alpha waves related to mental relaxation and concentration in young adult males.
A clinical study was conducted to assess whether l-theanine can modulate aspects of brain function in humans at realistic dietary levels. Electroencephalogram (EEG) was measured in healthy, young participants at baseline and 45, 60, 75, 90 and 105 minutes after ingestion of 50mg L-theanine (n=16) or placebo (n=19). Participants were resting with their eyes closed during EEG recording. There was a greater increase in alpha activity across time in the L-theanine condition (relative to placebo (p+0.05). A second study replicated this effect in participants engaged in passive activity. These results indicate that L-theanine, at realistic dietary levels, has a significant effect on the general state of mental alertness or arousal.
Melatonin is a hormone secreted by the pineal gland in the brain, well known for inducing and regulating sleep. Melatonin supplementation is known to normalize abnormal sleep patterns by decreasing sleep latency. It is one of the most widely used sleep aid supplements because it is effective and non-addictive. Melatonin also has general neuroprotective effects, can improve eye health and mood.
A series of 10 randomised clinical trials were conducted in airline passengers, airline staff or military personnel given oral melatonin, compared with placebo or other medication. The objective was to assess the effectiveness of oral melatonin taken in different dosage regimens for alleviating jet-lag after air travel across several time zones. Nine of the ten trials found that melatonin, taken close to the target bedtime at the destination (10pm to midnight), decreased jet-lag from flights crossing five or more time zones. Daily doses of melatonin between 0.5 and 5mg are similarly effective, except that people fall asleep faster and sleep better after 5mg than 0.5mg. Doses above 5mg appear to be no more effective. The results suggested that a short-lived higher peak concentration of melatonin works better than a small slow-released dose. The timing of the melatonin dose is important: if it is taken at the wrong time, early in the day, it is liable to cause sleepiness and delay adaptation to local time. The incidence of other side effects is low. The study concluded that melatonin is remarkably effective in preventing or reducing jet-lag, and occasional short-term use appears to be safe.
A study examined the induction of polysomno graphically recorded sleep by similar doses of melatonin given later in the evening, Volunteers received oral doses of melatonin (0.3 or 1.0 mg) or placebo at 6, 8, or 9 PM. Either dose given at any of the three time points decreased sleep onset latency and latency to stage 2 sleep. Melatonin did not suppress REM sleep or delay its onset. Most volunteers could clearly distinguish between the effects of melatonin and those of placebo when the hormone was tested at 6 or 8 PM. Neither melatonin dose induced "hangover" effects, as assessed with mood and performance tests administered on the morning after treatment. This study provides evidence that nocturnal melatonin secretion may be involved in physiologic sleep onset and that exogenous melatonin may be useful in treating insomnia.

I've always been sleeping poorly. It takes me forever to fall asleep, I wake up countless times a night. In the morning I feel like someone chewed and spitted me.

@xtyler i think its not a problem to use amnes1a in the evening if i already used re1gn for the training in the morning, right?
im asking because i would have 50mg kannaease in total and 25mg are 100% safe to consume daily, as i can see in the write up.
im extremely curious how i will react to amnes1a. i also wake up several times and the sublingual melatonin helps with the time to fall asleep but i need something that keeps me sleeping. the product looks outstanding, design is the best of your new products imho and the ingredients seem to be chosen very wisely, as always with OL products.

im just thinking of how i can get amnes1a without custom issues. in europe, or at least in my country, melatonin isnt really allowed to be selled in any amount. i think it is allowed to be selled in very small amounts in some products but its a little bit of a grey area. if i order from the US, i dont know, if customs pick it up. i didnt have any problems with the US-version of re1gn though.
so will there even be any intro sales on amnes1a as with your other products?
if so, i would want to buy 2 or 3 bottles because money is tight at the moment. but im not sure of the risk. maybe here are some guys who have successfully imported melatonin products from the states into europe?
if not, i can buy it anyhow out of UK or so because i wouldnt save that much money ordering from NV without a nice sale.

Hmm.. I thought the optimal melatonin dose was on the lower end towards the micrograms range? I've had sleep disruptions from 5 mg and I take 1 mg nowadays.
Looks good though, just needed rutaecarpine and this would've been the PERFECT blend =D. Gonna pick some up after my next paycheck

We use 3mg because this dose happens to work best when used in tandem with the other ingredients of the formula
